First create the puzzle piece in Photoshop5.5 on a transparent background.
Then choose save for web.
Select GIF as the file type, turn on transparency and finally (here's the
bit that fixes your problem) select a Matte colour that is the same as the
background colour on which the pieces will be placed in the Flash movie.

Using this solution the file size is no larger than a standard GIF. Only use
PNG if you want the pieces to look good on variety of contrasting
backgrounds.
