Using the example "Spotlight Mask" that ships with Flash 4, you could add an additional layer on top of all the other layers.  Copy the Layer:Mask layer's frames into this new layer (I called it "Overlay of color").  Then change the color effect of the "Art:spotlight light" symbol in both key frames, giving it an alpha value of approximately 50% (change to suit your needs).  The result will be a blue colored spotlight.


I'm sure there are other ways to do this, but this is one example.
