For any reasonable performance, you only want the video to play "Direct to
Stage".

However... you can include a mask channel in a QT video. Open up the video
in Movie Player, menu Movie>Get Info. Set the drop down on the left to:
Movie Track, Set the dropdown on the right to: Mask then click "Set" and
point to a PNG or other graphic file with an alpha channel (you could use
Fireworks and create a new document and use "transparent" for the background
color).

Plays well in Director!
